The absolute "most comfortable guitar strap" made is made by Jimmie Sorrels. He makes them in a boomerang shape (not that exagerrated, but you get the idea) and the "bend" in the strap is where it goes across your shoulder. He puts a slight cutout at that point so 2 things happen that are WONDERFUL!
1) The strap edge doesn't rub against your neck! 2) You never have to "wiggle" the strap back and forth during a gig! The straps are built to fit YOU... and mine is wonderful. Over 25 years ago, I got a wide thin single layer black leather guitar strap for a Christmas present from my brother-in-law (who was one of my guitar mentors).
It is simply the most comfortable strap I've ever felt in my life. Very simple and unadorned. Simple, single layer leather but 4" wide. Extremely soft and supple, like cloth. I have that on my latest electric. The only age it shows is a little stretch at the hole but I use straplocks so that doesn't matter and isn't seen. I wish I could find more like this. Not sure this helps except to say "a weird off-brand might be what you're looking for"

I have a couple Franklins that are very comfy, but my next strap will be a Lakota Leather. They are the softest leather I have ever felt (bison). I have one for my mandolin made of elk, but they do not make the elk in guitar sizes. You can buy them on ebay, or their website, or from American Bluegrass in Catonsville, MD.

For acoustic, I like a suede strap. For electrics, since they weigh more, I use the Levy that Wade posted. Works great, even for heavy basses.

My favorite is an old Peavey strap that my wife got me about 20 years ago.
I am right fond of my Levy strap as well. The trick, IMHO, is to find the widest strap you can stand to distribute the weight, and backed with suede so it won't slip around too much.

Most comfortable guitar strap?
I own a bunch of guitar straps, and I find that wider strap are more comfortable. Foam or fleece padding is also advantageous, particularly with electric guitars that are comparatively heavy.
About half of my straps were included with my guitar purchases, free of charge. Of the straps I've purchased, almost all cost under $15, including my Martin "Owners Club" soft leather strap. But my favorite straps are tastefully decorated. I make my selections on beauty, not comfort. Glen

I prefer a wide leather strap, no padding or seams. There are other devices that are designed to promote comfort, and I'm sure they do. But they also look and function like harnesses used by window washers and NASCAR drivers. Plain, wide leather gets it done.
