With your goal of a high end ust setup, and already owning the Avalon U5:
I’d suggest getting your ust connected directly to the U5 (bypass the DI and preamp in your guitar).
Make sure your UST is installed best it can be.
Get an interface with very good converters and if you might add a mic(s) in the future very good preamps. You should get an interface with line level inputs (that totally bypass its internal preamps) to connect to the U5 output. Another option is an external ADC which goes between the U5 and a digital input on an interface. You could also later add an external preamp for mics.

It helps to clarify your goal. Everyone has different tastes. I assumed you were hoping the U5 would help you get a more natural acoustic sound. With that John Mayer example, I guess the question is "why isn't your pickup into your interface getting you that sound?". It sounds very quacky to me, and I'd think you'd get a sound pretty close to that with your pickup right into the Scarlett (as long as you play like JM). Keep in mind it's a recording, and you don't know what was done in the mix. Could be a fair bit of compression, maybe a bit of saturation to make it a bit more gritty. Just a guess, based on a very quick listen.
