With the Fishman SA220, you have various choices and which one you pick will depend on what you need. If you've got a small mixer with effects, you want to use those effects instead of those on the SA220, and you're not connecting your SA220 to another SA220, your best choice is to go into the Monitor Input from one of the main outs of the mixer. This gives you a balanced connection that expects a line level signal, and it goes directly to the master output section of the Fishman. It basically allows you to use the SA220 as a powered speaker.
The other option for a balanced connection--and necessary if you want to use the effects and EQ on the SA220--is to go from a main output of the mixer into one of the mic inputs on the channels, but you'll need to be careful about your gain staging, since those inputs are expecting a mic level signal. Engaging the 10db pad might help. Or you can use an external pad like kingston suggests or even a passive DI box. You can use the instrument input, but it won't be balanced, although for short or even moderate cable runs that might not be a problem.
If you don't need the effects and EQ, but you do need to connect your SA220 to another SA220 using the monitor in/out connections, you can use either the Aux input or either one of the effects returns on the channels to send an unbalanced signal from your mixer to the SA220 using a simple 1/4" instrument cable. These will all insert your signal before the Monitor in/out connections, but after the unit's preamp/EQ/effects stages. Again, not balanced, but for short or moderate cables probably OK.
If you need to send a stereo signal to the SA220 so it can sum it to mono (if, for example, you're running a stereo keyboard or some other stereo signal into your mixer and your mixer does not have its own mono output), you can use the Aux input, which is a stereo TRS input that will do the summing for you. Use a stereo Y cable with whatever connectors you need on the separate ends (1/4", RCA, XLR etc.) and 1/4" TRS on the end that goes into the SA220.
Or you can send the left and right main outputs from the mixer to the two channel inputs and mix them that way.
