I'd be curious to understand how much a truss rod affects tone and/or sustain.

I'm thinking compared to a solid neck, a truss rod, especially a 2 way one will have an impact on tone even if only by virtue of having removed some wood from the neck to make room for the rod. Then of course the weight and mass added back by the steel may have influence as well.

Would it help or hurt if the true rod were made of lighter material than steel?

As far as I can see the steel truss rod would lower the resonance of the neck and add sustain. How much if any? Might be time for me to make up a neck to experiment with.
